Job description

Early Stage Researcher 14: Nanocarriers for combined delivery of siRNA and low molecular weight drugs (LMWD) to the peritoneal cavity

You will perform your PhD research in a multidisciplinary Innovative Training Network (ITN) project called NANOMED. The aim of this particular PhD project is to search for biocompatible nanocarriers suitable for intraperitoneal siRNA delivery and to further improve their performance via repurposing of LMWDs. The task of the ESR is to develop and evaluate a series of nanoformulations for the combined encapsulation and administration of siRNA and LMWDs to the peritoneal cavity. High encapsulation efficiency of siRNA and LMWDs should be obtained, with little to no premature siRNA release in biological extracellular media such as blood and intraperitoneal fluids. Biological activity, colloidal stability, toxicity and in vivo biodistribution of selected formulations will be evaluated. Nanocarriers with good in vitro stability in biofluids will be tested in animal models with induced peritoneal carcinomatosis by SKOV-3 cells expressing firefly luciferase. The formulations will be injected or nebulized intraperitoneally and their in vivo stability will be evaluated as a function of time by performing advanced microscopy techniques on samples taken from the IP fluid and the blood. Ideally, the carriers should not aggregate and remain in the peritoneum for a sufficiently long time. The biological activity of the nanocarrier formulations will be assessed by quantifying downregulation of firefly luciferase and tumor growth.

What we have to offer:

• employment: 1,0 fte;

• tax free fellowship including full social security coverage (net monthly amount starting at ± 1.900 EUR/month plus end-of-year and vacation allowance), mobility allowance and (if applicable) family allowance

• duration of the contract: 1 year, with option to extend

Profile of the candidate

At the time of recruitment you must be in the first four years of your research career and have not yet been awarded a doctoral degree. You have excellent communication skills and are able to work efficiently in a team. Recruitment is open to all nationalities, but because of mobility rules defined by the EU, at the time of recruitment, candidates must not have resided or carried out their main activity (work, studies, etc.) in Belgium for more than 12 months in the three years immediately prior to the reference date (compulsory national service and/or short stays such as holidays are not taken into account).
